Preparation Of 7% Bsa/5 Mm Cold Palmitate Mixture
BSA is required to act as a carrier for palmitate to ensure lipid solubility in an aqueous solution. Cold palmitate is included in the FAO reaction in order to increase the total rate of oxidation and signal measured in the assay. In order to prepare a solution of 7% BSA/5 mM palmitate , do the following:
Prepare a 7.5% BSA solution by diluting a 30% stock in ddH2O. Warm to 42 Â°C in a water bath. Prepare 20 mL for the 7% BSA/5 mM palmitate solution and 20 mL for a 7% BSA only control solution.
Weigh 27.8 mg sodium palmitate and put in a 50-mL conical tube. Add 1.3 mL of ddH2O and keep cap closed but loosened.
Place the tube in a boiling water bath until the fatty acids are dissolved .
Cool the palmitate until it can be held in a bare hand, but the fatty acids are still dissolved . If the palmitate precipitates, then warm again.
Immediately add 18.7 mL of the 7.5% BSA. Place in a 42 Â°C water bath for 30 min. For the 7% BSA only control, add 18.7 mL of the 7.5% BSA to 1.3 mL of ddH2O.
If particles form and are visible to the naked eye, sonicate for 5 min. Increase the temperature to 47 Â°C if particles are still present, but do not go above 50 Â°C to avoid denaturing the BSA.
Store unused aliquots at â 80 Â°C for no more than 6 months. Avoid freezeâthaw cycles.

What Is A 1% Solution
The word solution describes something that exists as a liquid. For example, both water and milk are solutions. When we say that a solution of water is 1% sugar, it means that 1% of the molecules in that solution are sugar molecules, while the other 99% are water molecules. Thus, a 1% solution of BSA means that 1% of the molecules in that solution are BSA. Since BSA is often sold as a dry powder, a certain amount of it needs to be weighed out and then dissolved in a liquid, such as water. Dissolving BSA powder into a liquid is called a weight-by-volume dilution, often written as w/v in the instructions that accompany chemicals or scientific experiments.

Beginning in the fall semester of 2014, students in the College of Natural Sciences will be able to earn a new degree, the Bachelor or Science and Arts degree. Its a cross-disciplinary degree for those students interested in combining a core science experience with course work in the fine arts, liberal arts, communications, or business. Students should speak to their advisors for the more fine-grained details of the new degree, but to get the bigger picture, I sat down for a few minutes with Dr. Sacha Kopp, associate dean for undergraduate education.
Daniel Oppenheimer: Why the BSA?
Sacha Kopp: Most of the students in this college follow a degree plan called the Bachelor of Science, or BS, which is effectively preparing you to be a specialist in that field. Its a great degree plan if youre going to graduate school in a given science or if youre going to be a specialist. But we have 11,000 students in this college, and they have a lot of diverse interests. A lot of them want to explore science and math but they want to use it in some interdisciplinary way. We needed a better degree structure to support that.

Youve probably heard of BMI in reference to overall health, weight loss or exercise, but what about BSA ? How is this different from BMI and how is it calculated? Moreover, why should you care?
While Body Mass Index is a measure of a persons body fat mass, Body Surface Area measures the total surface area of a persons body and is frequently used in order to calculate drug dosage and the amount of fluids to be administered by IV.
Various formulas have been used over the last century to calculate BSA. They all give slightly different results, which has created issues of standardization. For the sake of example, the Mosteller formula is commonly referred to and often used in clinical trials. It calculates BSA by taking the square root of height multiplied by weight divided by 3600.
Many online calculators offer the conversion from centimeters to inches and from kilograms to pounds, which makes calculating ones own BSA fairly easy. Take, for instance, a woman who is five feet, 5 inches tall and who weighs 150 pounds. This calculator allows you to select inches and pounds and the type of formula you want to use. Youll see that the woman in this example has a BSA of 1.75 m2 using the DuBoise formula.
It is also routinely calculated in the cardiovascular field, and research has shown that body surface area is a strong predictor of mortality in chronic heart failure patients.

Several BSA-based âneoglycoproteinsâ were shown to stimulate the acrosome reaction. The binding of Man,Flu-BSA results in a time-dependent receptor aggregation and the induction of acrosome exocytosis in capacitated sperm populations from fertile donors.200N-acetyl-Î±-glucosamine and Î±-mannose may interact with the putative receptor for one protein of zona pellucida: ZP3 in human spermatozoa.195,202â205 This binding results in the exocytosis of the sperm acrosome .206 The GlcNAc-BSA-induced acrosome reaction is inhibited by a small ligand: N-acetylglucosamine and by a purified soluble hydrolase, Î²-N-acetylglucosaminidase. The induction of the AR with Man-BSA was inhibited by mannose, while soluble Î±-mannosidase was only partially effective. These data suggest that binding sites for GlcNAc and Man seem to be involved in the induction of the AR in human sperm. The induction of AR in human spermatozoa by GlcNAc-BSA could be used to predict their fertilizing ability in vitro.205
The effect of âperitoneal fluidâ on the human sperm acrosome reaction was tested.208 When the AR was induced by GlcNAc-BSA, pre-incubation with peritoneal fluid reduced the percentage of AR, while peritoneal fluid from either the endometriosis group or infertile patients without endometriosis caused no significant differences. These data indicated that peritoneal fluid possesses a protective factor which prevents premature AR.

Nope. Its not the same. Fetal Bovine Serum is a commonly used serum supplement for eukaryotic cell culture. The benefit of FBS for cell culture is its lower antibody levels and higher growth factor levels. Bovine Serum Albumin is used in a variety of laboratory applications including its function as a protein concentration standard, its function as a cell nutrient and its ability to stabilize enzymes during restriction digest. The important takeaway here, however, is that while they often both come up in Google searches when youre only looking for one or the other, and sound very similar, they are indeed different.

The Bovine Serum Albumin Standard
The ideal protein standard for protein quantification measurements is a purified sample of the protein youll be assaying in your unknown samples. However, that may not always be an option. If a purified sample is unavailable, or is too costly, then youll need an inexpensive and readily available alternative enter bovine serum albumin.
Bovine serum albumin is a globular protein that is often used as a protein concentration standard in lab experiments as well as in numerous other biochemical applications. Derived from cows, BSA is extracted from cow blood using one of three different purification methods: cold-organic solvent fractionation, heat shock, and ion exchange chromatography.
It is very useful in different biochemical and life science applications because of its lack of effect in many different biochemical reactions as it does not affect other enzymes that do not need it for stabilization. Another useful aspect of bovine serum is that it is small and can be easily stored. It is stable at room temperature for up to three weeks. In a solution, bovine serum should be stored between 2 and 8 degrees Celsius .
Finally, BSA offers features like sensitivity, compatibility with common substances in samples , standard curve linearity, and protein-protein variation for reliable, consistent results.

BSA may protect mammalian cells against certain genotoxic agents . Genotoxic agents are mutagenic chemicals that damage cell DNA. This can result in mutations or cancers.

Weight Percentile And Height Percentile Compared To The Population
An adult whose weight is at the 50th percentile, is at the average weight of the population. An adult at the 90th percentile is quite heavy.
For calculating percentiles, the oldest version of the calculator used weight data from a study9 of 3992 consecutive adult cancer patients who underwent CT scanning. That study did not stratify males versus females, and it did not include pediatrics. By using the variance distribution from that study, and then adjusting the means to 79.4kg for males and 64.5kg for females, it was able to estimate the weight percentile. The height data came from a study10 of 600 randomly selected cancer patients. That study also did not stratify males versus females, and it did not include pediatrics. By using the variance distribution from that study, and then adjusting the means to 69 inches for males and 64 inches for females, the older calculator was able to estimate the height percentile.
The next version of the calculator used better data. Pediatric data was derived from standard pediatric growth charts, based on American 1979 data13. Adult data for median height and weight came from Canadian 1997 data11. The variances for height and weight and the weight for height data comes from Canadian 1971 data12. Americans tend to be a little taller and heavier than Canadians.

BSA powder is very light and is often used in small amounts, so researchers need a very sensitive weighing scale in order to accurately measure out BSA. Researchers often do not need large amounts of a BSA solution, so making several gallons or liters at once would be a waste of powder and storage space. Biochemistry and molecular biology experiments often use small amounts of BSA solution, and it is generally good practice to make it fresh each time. A simple way of making a 1% BSA solution is to weigh out one gram of BSA powder, pour it into a graduated cylinder that can hold more than 100 milliliters of water, and then add water until the liquid level reaches the 100 mL mark. Mathematically, one divided by 100 equals 1%.

Bovine Serum Albumin , also known as “Fraction V“, is a protein derived from bovine blood plasma and is a byproduct of the food industry. The nickname “Fraction V” is based on the plasma protein extraction method by Cohn, which makes use of the different solubility behavior of the plasma proteins. In this fractionation, BSA represents the fifth fraction and is based on cold ethanol precipitation.
In order to yield the mature BSA protein, the full-length precursor protein is modified in two steps: First, a signal peptide is cut off upon secretion. During the second step that yields the mature BSA protein an additional 6 amino acids are removed. The final BSA product then contains 583 amino acids.
